Cancer stem cells (CSCs) drive tumor growth and are responsible for tumor resistance to therapy. Here, we report that lung CSCs are characterized by higher stemness and drug-resistance properties than their differentiated progeny. We show that CD44+ lung CSCs exhibit high expression of N-linked glycosylation genes and that CD44+ lung CSCs are more resistant to chemotherapeutic drugs than CD44+ cells. Using a glycosylation inhibitor and shRNA against glycosyltransferase genes in CD44+ lung CSCs, we show that glycosylation of CD44 is critical in maintaining stemness. In addition, using glycosidase inhibitors, we show that lysosomal targeting plays a major role in the maintenance of stemness in CD44+ lung CSCs.Conformational analysis of dioleoyl-trimethylammonium bromide molecules in the bilayer phase by conductance measurements.
The conductance and capacitance measurements were used to obtain structural information about dioleoyl-trimethylammonium bromide (DOTAB) bilayer membrane. The electrical measurements of individual DOTAB bilayer at different temperatures were used to calculate the potential barrier profile using the Poisson-Boltzmann model. The simulated bilayer was found to be in good agreement with the experimental results. These measurements also indicate the existence of various structural changes in the DOTAB bilayer membrane at different temperatures. The results presented here indicate that the peak-to-peak height is approximately 0.65 mV at 30 degrees C. A decrease in the peak-to-peak height of 0.37 mV was observed when temperature was increased to 35 degrees C.
